【摘要】:Mycoplasmacontaminationofcellculturesisacommonproblemthatcanaffectthegrowth,metabolism,andfunctionofculturedanimalcells.Theabilitytodetectmycoplasmaincellculturesprovidesanopportunitytoensurethatcellsarefreeofcontamination,andtoreplacethosethatarenot.Foradditionalinformation,seePracticesE1531,E1532,andE1536.Strictadherencetoestablished,well-testedproceduresisnecessary.ThispracticewasdevelopedbyTaskGroupE48.01.02toassistindevelopingandmaintaininganestablishedregimenformycoplasmadetectionbyindirect4x2032;-6-Diamidino-2-Phenylindole(DAPI)fluorochromestaining.Thispracticeisintendedforuseinexaminingculturedanimalcellsforthepresenceofmycoplasmacontamination.Thispracticeisnotintendedforuseinthedetectionofmycoplasmacontaminationinserum,culturemedia,orsystemsotherthanculturesofanimalcells.Allcellculturestobeexaminedformycoplasmashouldundergoaminimumoftwopassagesinantibiotic-freetissueculturemediumbeforetesting.1.1ThispracticecoversproceduresusedforthedetectionofmycoplasmacontaminationbyindirectDNAstaining.1.2ThispracticedoesnotcoverdirectmethodsforthedetectionofmycoplasmaorotherindirectmethodssuchasenzymaticaldetectionorDNAprobes.1.3Thispracticedoesnotcovermethodsfortheidentificationofmycoplasmaorganisms.1.4ThevaluesstatedinSIunitsaretoberegardedasthestandard.Thevaluesgiveninparenthesesareforinformationonly.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
